Cytochrome-P450-CYP27B Definition
Encoded by human CYP27B1 Gene (Cytochrome P450 Family), 508-aa 57-kDa (precursor) Cytochrome P450 CYP27B is a kidney inner mitochondrial membrane P450 heme-thiolate monooxygenase that hydroxylates 25-hydroxyvitamin D3 at the 1alpha position to synthesize (second, last step) 1alpha,25-dihydroxyvitamin D3, the active form of vitamin D3 that binds to vitamin D receptor and regulates calcium metabolism. In regulating the level of biologically active vitamin D and calcium homeostasis, CYP27B1 plays an important role in normal bone growth and tissue differentiation. CYP27B1 defects cause vitamin D-dependent rickets type 1 (VDDR-1). P450 enzymes catalyze reactions involved in drug metabolism and synthesis of cholesterol, steroids and other lipids. (NCI)
