EWS-FLI1 Type 1 Definition
One allele of the transforming chimeric gene resulting from a t(11;22) translocation that is found frequently in patients with Ewing's sarcoma or primitive neuroectodermal tumors (PNET). EWS-FLI 1 type 1 encodes a protein, mTOR, that is a central regulator of translation and cell proliferation. mTOR activates the mitogen-activated protein kinase (MAPK) signaling pathway, resulting in neoplastic transformation. Antisense oligonucleotides that target EWS-FLI 1 type 1 have been used investigationally to treat this genetic anomaly, resulting in cell cycle arrest at G1 phase. (NCI04)
