Growth Hormone Receptor Definition

Widely expressed by human GHR Gene (Cytokine Receptor I Subfamily 1), Growth Hormone Receptor is a type I membrane receptor for growth hormone with a ligand-binding extracellular domain (GHBP); a FnIII domain; a WSXWS motif; a cytoplasmic UbE motif for ubiquitination and internalization; and a cytoplasmic JAK interaction box 1 motif. Produced by cleavage by cell surface ADAM17, secreted GHBP may modulate GH signaling; GH appears to inhibit ADAM17. Tyr phosphorylated on GH binding by JAK2, homodimerized GHR appears to activate JAK2/STAT pathways that produce IGF1, which activates mitogenic and anabolic growth responses. SOCS3 may inhibit JAK2 activation while CIS and SOCS2 may inhibit STAT5 activation by GHR. GHR defects cause Laron dwarfism and idiopathic short stature. (NCI)
