Vinculin Definition

Encoded by a single human tumor suppressor VCL Gene, Vinculin contains 1,066 amino acids with strong similarity to other vinculins and alpha-catenins. Alternative splice product metavinculin contains additional 68-79 variable and highly conserved amino acids near the C-terminus. Involved in cell adhesion, vinculin is associated with the cytoplasmic face of adherens-type junctions and involved in anchoring F-actin to the plasma membrane. Vinculin is phosphorylated on serines, threonines and tyrosines and is acylated by myristic acid and/or palmitic acid. Vinculin exhibits self-association properties and appears to be the major talin-binding protein in platelets; 150 kD metavinculin also interacts strongly with talin.
